Let's start a new thread for links to Game Over videos out there. There are a bunch on Youtube (and other sites too).
Since we haven't yet had the opportunity to make our own video, fan works like this are just incredibly cool.

That's some great shit!  :D You've got to watch this a couple of times though to really get it, seems like there's a lot that flashes by.

Extract the maps, record a playthrough. Merge the two.
Record the characters position at various points (given that they didn't have a computer controlled rail-mounted camera they couldn't get that part perfect. You'll note that the camera doesn't follow mario perfectly).

Then you find a suitable location, calculate the length of the maps and note the positions where your character was at different points in time. Use that data to move the camera along from point to point, following the previously mentioned schedule.

The final part is merging the two through the use of fairly simple shape recognition software (recognize the shape upon which you're suppose to project the digital image. Pretty simple in this case). Tweak until you're satisfied.

Simple in theory, but requires a lot of work.

Not really. If I was going to do a project like this I wouid probably pick Mario as well, and despite being a gaming freak Mario never really appealed to me.
I've finished pretty much all the old school mega man (and sonic and a dozen other plaformers) games, but I've never taken the original Mario beyond level 10-ish. I've never finished Super Mario World (although I've gotten to the star levels).

I'd pick it because it's recognizable (public appeal), the levels are fairly flat (unlike Sonic or Megaman which have a lot more lateral movement. A horizontal level layout makes it simpler for the face recognition software to fix the image upon the selected medium), low-detail background (cleaner projection) and bright/clean colors (again, cleaner projection).

Something people refuse to believe is that the guy DOES know how to sing, it's just that in those parts, he sings like that on purpose. If you listen to other of his songs, you'll notice.
